Proteomics is the analysis of the entire protein complement of a cell, tissue, organism, or species. The application of technologies to identify and quantify proteins is crucial in early disease diagnosis, prognosis, and drug development.

A support or matrix in affinity purification is any material to which a biospecific ligand is covalently attached. affinity chromatography, or SMOAC (Figure 1B), in which phosphopeptides are first enriched by TiO 2 resin followed by enrichment with Fe-NTA resin.

Affinity proteomics

Affinity depletion is based on the specific capture of target proteins using immobilized antibodies or other molecules (protein A/G, Cibacron Blue) with high affinity and specificity for the targets. Simultaneous removal of multiple high abundant proteins has become the major pre-analytical strategy for proteomic analysis of blood plasma [ 9 ].

For this, we applied affinity proteomics, utilizing tandem affinity purifications combined with mass spectrometry. Analysis of the acquired proteomics data provides evidence that ADGRs not only have functional roles at synapses but also at intracellular membranes, namely at the endoplasmic reticulum, the Golgi apparatus, mitochondria, and Keilhauer et al.
